Amyloid beta-peptide(25-35) （AbMole，M9071）作為全長β-淀粉樣蛋白（Aβ1-42）的功能片段，在阿爾茨海默病（AD）的研究中被廣泛應(yīng)用于細(xì)胞和動物模型。在動物模型中，β-Amyloid (25-35)（CAS No.：131602-53-4）一般要在pH為7.4的生理鹽水中孵育36小時以誘導(dǎo)聚集，然后通常通過立體定向注射的方式進(jìn)行給藥處理，例如有文獻(xiàn)通過雙側(cè)注射的方式直接注入大鼠海馬CA1區(qū)域，注射濃度為0.1 µg/µL，注射體積為1μL，這種處理方式能夠顯著改變海馬區(qū)的樹突棘形態(tài)，并影響與空間記憶相關(guān)的NR2B和PSD-95蛋白的表達(dá)[1]。在另一個AD大鼠模型中，Amyloid beta-peptide(25-35) （AbMole，M9071）通過立體定向注射直接注入海馬區(qū)，劑量為6.25 µg（單次注射）[2]。一般在Aβ25-35注射后8周內(nèi)，動物表現(xiàn)出顯著的記憶和空間導(dǎo)航障礙，此時可通過Morris水迷宮、Y迷宮等行為測試驗(yàn)證[3]。或者通過病理學(xué)檢測分析動物腦部組織的變化，如海馬區(qū)β-淀粉樣蛋白沉積、糖原合成酶激酶-3β（GSK-3β）激活、AKT信號抑制等[4]。在細(xì)胞實(shí)驗(yàn)中，Aβ25–35 可以25 μg/ml的濃度誘導(dǎo)海馬神經(jīng)元凋亡[5]，或者以2.5-10μg/mL的濃度誘導(dǎo)角細(xì)胞的凋亡[6]。
